      Theme: 
      Plastic pollution is a major challenge for our generation, and plastic recycling can both reduce pollution and limit the fossil-based production of new plastics. Enzymes able to break down plastics could sustain such a circular economy. Polyethylene terephthalate (PET)-degrading enzymes (PETases) have attracted considerable interest and have been extensively engineered to improve their performance.
      Since 2022, in collaboration with Carbios, a leading biotech company in enzymatic recycling, we have been studying by NMR one of the most efficient PETases, LCC-ICCG, a quadruple mutant of Leaf-branch Compost Cutinase. In this seminar I will present several complementary approaches to measure the pKa values of histidines residues in the PET-degrading enzyme LCC-ICCG. I will show how these values can be linked to catalytic activity, and why it is essential to measure them on the proper nucleus to obtain a meaningful picture of the catalytic mechanism. In the second part, I will discuss how NMR can provide residue-level insight into the factors that govern protein stability, and how this information could guide the rational design of more thermostable enzymes.
