Characterize macromolecule samples by 1D, 2D NMR, recording of 2D, 3D, nD NMR experiments and assignments.
MW. Below 15 kDa

The sample quality is checked by 1D/2D NMR, then 3D or nD NMR experiments are recorded, and spectra are assigned.
Output : Assignment File in BMRB Format.

Structure Determination

Goal

Recording of 2D/3D NOESY NMR spectra, automatic assignments using Cyana 3 and refinement.
MW. Below 15 kDa

The structure is calculated using unassigned NOESY peak lists, assigned chemical shifts list and dihedral constraints. The preliminary structure is refined step-by-step.
Output : Assignment File in BMRB Format. Structure in PDB format.

HP NMR and screening

Goal

Folding pathway of proteins at the residue level. Ligand screening at low throughput.
MW. Below 15 kDa for HP NMR

Series of HSQC spectra are recorded at increasing hydrostatic pressure.
Thermodynamic analysis allows characterizing the unfolding/folding pathway.
Output : Series of thermodynamic parameters and intermediate structures.
